Short answer: most do not, but "do testosterone boosters work" is the wrong question, & that is why it has never had a good answer online. The same ingredient can produce a measurable result in one group of men & nothing at all in another. The useful question is narrower: which ingredient, for which outcome, in which population.

A 2023 systematic review in the International Journal of Impotence Research examined 52 studies covering 27 proposed testosterone boosters, & did something almost no consumer article does. It analysed four populations separately: male athletes, men with late-onset hypogonadism, infertile men, & healthy men with normal levels. The results diverge sharply between them.

What follows is a verdict on each of the common ingredients, with the trials attached. Which Ingredients Actually Have Evidence

Four ingredients clear the bar. The rest either fail, or work only in specific circumstances that the label never mentions.

What Counts as a Testosterone Booster?

A testosterone booster is any over-the-counter supplement marketed on the claim that it raises the body's own testosterone production. It is a marketing category, not a pharmacological one, which is the root of most of the confusion.

The category contains vitamins, minerals, amino acids, botanical extracts & combinations of all four. Nothing unites them chemically. The only thing they share is the claim on the label.

This matters because it makes any blanket verdict meaningless. Asking whether testosterone boosters work is like asking whether "things sold in bottles" work. A 2019 analysis published in the Journal of Sexual Medicine, examining the best-selling testosterone boosters on Amazon, found limited human research behind them & no definitive findings of efficacy for the category as sold.

Testosterone boosters are also distinct from testosterone replacement therapy. TRT introduces exogenous testosterone under medical supervision for diagnosed hypogonadism. A supplement attempts to nudge your own production. They are not comparable, & no supplement should be presented as an alternative to a diagnosis.

Why the Same Ingredient Gives Different Answers

The single most useful finding in this field is that results depend on who is taking the supplement. The 2023 systematic review structured its entire analysis around this, reporting outcomes separately across four groups.

There is a straightforward physiological reason for this. A supplement that supports a pathway can only produce a visible change if something in that pathway is underperforming. In a man whose testosterone is already comfortably normal, there is little headroom. In a man with a deficit, there is.

Almost every argument about testosterone boosters online is two people describing different populations & assuming they are describing the same thing.

Worth sitting with

This is why a blood test is worth more than a supplement decision. If your testosterone is mid-range & your symptoms come from sleep debt or training load, no ingredient in this article will do much. If it is genuinely low, that is a medical conversation. Either way, the test comes first.

Ingredient by Ingredient: What the Trials Found

Ashwagandha (Withania somnifera)

The strongest evidence of any common herbal ingredient, though the effect is modest. A systematic review published in Advances in Nutrition in 2021 examined four randomised, double-blind, placebo-controlled studies covering 197 men. Three of the four found that at least eight weeks of ashwagandha raised testosterone more than placebo. One found no effect.

The most-quoted individual trial (Wankhede et al., 2015) reported a mean testosterone increase of 96.2 ng/dL against 18.0 ng/dL for placebo over eight weeks. An important caveat that most summaries omit: every participant was also following a resistance-training programme, & resistance training raises testosterone independently. The trial shows ashwagandha added to that effect. It does not show what ashwagandha does on its own.

Ashwagandha's better-established action is on stress & sleep, which matters here because sustained high cortisol suppresses testosterone. We covered the comparison in more depth in Ashwagandha vs Shilajit.

Shilajit (purified)

The single cleanest testosterone result in this article, from one trial that nobody has repeated. A randomised, double-blind, placebo-controlled study published in Andrologia in 2016 gave healthy men aged 45 to 55 either 250 mg of purified shilajit twice daily or placebo for 90 days. Ninety-six enrolled, 75 completed. The shilajit group showed roughly a 20% increase in total testosterone against placebo, with increases in free testosterone & DHEA-S alongside it.

Two design details make it stronger than most trials in this field. It ran for 90 days rather than the four to eight weeks typical of supplement research, & luteinising hormone & FSH stayed stable, which suggests the effect did not come at the cost of disrupting the signalling chain above the testes.

The caveat is straightforward. It is one trial, in one narrow age band, in healthy volunteers rather than men with diagnosed low testosterone, & it has never been replicated at scale. That makes it the most promising single result in this article rather than a settled one.

The 2023 systematic review reached a similar position by a different route. Assessing a standardised purified shilajit extract, it placed the ingredient in its "possibly effective" category for men with late-onset hypogonadism - a cautious grade, but one awarded to only a handful of the 27 substances it examined.

One practical caveat that matters more with shilajit than with any other ingredient here: the trials used purified extract, & adulteration in the retail market is common. We covered dosing, timing & what home purity tests can & cannot establish in how to take shilajit.

Ashwagandha & shilajit are the only two ingredients here with testosterone-specific human evidence worth the name, & they act through entirely different mechanisms: one on the stress axis, the other on cellular energy production. That difference is the actual argument for taking them together, & it is why Ayamveda sells them as a pair in Shilajit Resin + Vitality Mix rather than as competing options. Note that shilajit is not an ingredient in the Gym Edition formula discussed later.

Tribulus terrestris (Gokshur)

Does not reliably raise testosterone in men with normal levels - but the evidence for sexual function is better than its reputation suggests. This ingredient gets its own section below, because it is the clearest example of why the population question matters.

D-aspartic acid

The most striking negative finding in the category. A 2015 trial published in the Journal of the International Society of Sports Nutrition gave 24 resistance-trained men either 3g of D-aspartic acid, 6g, or a placebo. The 6g dose reduced total & free testosterone relative to baseline. The 3g dose produced no significant change in either.

The sample was small, & earlier work in untrained men had suggested a positive effect. But a supplement sold specifically to raise testosterone producing the opposite result at a higher dose is worth knowing about, & it almost never appears in consumer coverage.

Vitamin D, zinc & magnesium

These correct deficiencies. Correcting a deficiency is not the same as boosting. The 2023 review covered ten studies of cholecalciferol & five of zinc or magnesium, & found they generally fail to raise testosterone in men who are not deficient to begin with.

That is not an argument against them. If you are deficient in vitamin D, fixing that is worth doing for reasons well beyond testosterone. It is an argument against expecting a supplement to do something for you that it can only do for someone with a shortfall you do not have.

Creatine

One of the best-supported supplements in sports nutrition, & not a testosterone booster. The evidence for creatine on strength, power & lean mass is extensive. The 2023 review found it among the ingredients that often fail to significantly increase testosterone. Both things are true, & creatine does not need the testosterone claim to justify itself.

Betaine & HMB

The 2023 review named betaine & beta-hydroxy beta-methylbutyrate among the few ingredients showing an effect, specifically in male athletes. Worth noting the qualifier: that finding applies to a trained population, not to the general market these products are sold into.

Tongkat Ali (Eurycoma longifolia)

Better supported than its profile in the West suggests. The 2023 review counted three studies & rated Eurycoma longifolia possibly effective in two of its four populations: men with late-onset hypogonadism, & healthy men. Alongside ashwagandha, it is one of only two botanicals the review flagged as possibly effective in healthy men specifically.

Three studies is a thin base, & "possibly effective" is a deliberately cautious grade rather than an endorsement. But it sits above most of this category, & it deserves more attention than it usually gets in Indian formulations, where it is rarely used.

Fenugreek, maca & the rest

The 2023 review counted two studies each of Lepidium meyenii (maca) & isoflavones, with most remaining ingredients represented by single reports. That is not enough to support a confident verdict in either direction, & anyone offering one is going beyond the evidence.

The Tribulus Case Study: Right Ingredient, Wrong Claim

Tribulus is the clearest illustration of why "does it work" is the wrong question. The marketing claim & the evidence point in different directions, & both camps in the argument are partly wrong.

On testosterone, the sceptics are right. A 2025 systematic review of clinical trials in Nutrients found no robust evidence that Tribulus raises testosterone in men with normal baseline levels. Eight of ten studies reported no significant change in androgen profile, & the review noted that participants in those neutral studies did not have low androgens to begin with.

On sexual function, the sceptics are wrong. A 2017 trial published in Maturitas randomised 180 men aged 18 to 65 with mild to moderate erectile dysfunction to Tribulus extract or placebo for 12 weeks. The Tribulus group improved on the International Index of Erectile Function by 2.70 points more than placebo (95% CI 1.40 to 4.01), with significant separation on intercourse satisfaction, sexual desire, orgasmic function & overall satisfaction. No drug-related serious adverse events.

A separate double-blind, placebo-controlled Indian trial in men with oligozoospermia found a significant advantage for Gokshur on a quality of sexual life measure, while the hormone changes between groups were not statistically significant. Two independent trials, same pattern: an effect on sexual function, no reliable effect on testosterone.

The distinction

Tribulus has reasonable human evidence for sexual function in men who have a problem. It does not have good evidence for raising testosterone in men who do not.

Both statements are supported. Marketing that quotes the first to justify the second is where this category goes wrong, & it is worth saying that Gokshur appears in our own formulation, where the honest description of its role is sexual function & desire support, not testosterone.

Can a Testosterone Booster Lower Your Testosterone?

In at least one documented case, yes. The D-aspartic acid trial above found that the higher of two doses reduced both total & free testosterone in resistance-trained men over the study period.

Beyond that specific finding, there are two structural risks worth understanding.

Proprietary blends. A proprietary blend lists ingredients without disclosing the amount of each, giving only a combined total. This is legal in most markets & it is the mechanism by which underdosing happens: a formula can list an impressive ingredient at a fraction of the trial dose & you cannot tell. If a label will not tell you the milligrams, you cannot compare it to any published study.

Adulteration. Supplement regulation is light in most markets, & analyses of products sold for male sexual health have repeatedly found undeclared pharmaceutical compounds. That is a genuine safety issue, & it is the argument for buying from manufacturers operating under real regulatory oversight.

What to Look For in a Formula

Since the category verdict is mostly negative, the useful skill is telling the defensible minority from the rest. Four criteria, in order of importance:

  1. Disclosed milligram doses for every ingredient. Not a proprietary blend. If you cannot see the dose, you cannot check it against a trial.
  2. Standardised extracts rather than raw powder. Raw botanical powders vary substantially in active-compound content between batches, which is why two people on nominally the same product can have different experiences. Standardisation means each batch is tested to a fixed percentage of actives.
  3. Ingredients with human trials, at doses that match them. An ingredient with promising rodent data at a dose nobody has tested in people is not evidence.
  4. Realistic timelines. Anything promising results in days is describing something other than hormonal change. Eight to twelve weeks is the honest window.

Applied strictly, those four criteria disqualify most of the market. The first one alone removes the majority: if a label gives you a blend total instead of per-ingredient milligrams, there is no comparison to make.

Single-ingredient doses versus blends

One honest complication, because it decides what you should expect. Almost every trial in this article tested one ingredient on its own, at a dose chosen to maximise the chance of detecting an effect - 300 to 600 mg of ashwagandha root extract, 250 mg of shilajit twice daily. A multi-herb blend distributes its total across six or eight ingredients, so each one sits below the single-ingredient trial dose.

Neither approach is automatically right. A single-ingredient product at trial dose gives you the best chance of the specific effect that trial measured. A blend covers more mechanisms at lower individual doses, which is closer to how Ayurvedic formulation has always worked - Rasayana preparations were layered deliberately, not isolated.

What matters is knowing which one you are buying, and that is only possible when the label discloses milligrams. If your priority is a single ingredient at the dose its trial used, the more direct route is a standalone product: purified shilajit at 250 to 500 mg sits squarely in the range the 90-day trial tested.

When to Skip Supplements & See a Doctor

If you have symptoms that suggest genuinely low testosterone, the correct first step is a morning blood test, not a purchase. The symptoms - fatigue, low motivation, reduced libido, stalled training progress, stubborn central weight gain - overlap almost perfectly with ordinary sleep debt, stress & overtraining, which is exactly why guessing from how you feel is unreliable.

See a doctor rather than a supplement aisle if:

  • Symptoms have persisted for months & are affecting daily function
  • You have lost morning erections consistently
  • You & a partner have been trying to conceive for a year without success
  • There are other signs of hormonal disruption, such as breast tissue changes or testicular shrinkage
  • You have diabetes, obesity or a thyroid condition, all of which affect testosterone directly

What decides your outcome more than any ingredient is whether you had room to move in the first place. Get a morning blood test if your symptoms suggest a problem. Fix sleep, training load & body composition, which move testosterone more reliably than anything sold in a sachet.

If, after all that, you still want to try something, the evidence points somewhere specific. Ashwagandha & purified shilajit are the only two ingredients in this article with testosterone-specific human evidence, they act on entirely different systems, & nothing suggests they interfere with each other. Taking both, at disclosed doses, for a full 8 to 12 weeks, is the version of this decision that follows the research rather than the marketing. Expect modest rather than dramatic. Judge it at twelve weeks, not three. And if nothing has shifted by then, stop - that conclusion is worth more than another jar.
